چکیده

Isolated photosynthetic reaction center from the bacterium Rhodospirillum rubrum was extracted with acetone-methanol. Its main pigments were identified as bacteriochlorophyll, bacteriopheophytin, and spirilloxanthin. The extinction coefficients of these pigments in acetone-methanol were determined. Quantitative spectroscopic analysis of the dry acetone-methanol extracts indicated a bacteriochlorophyll to bacteriopheophytin mole ratio of 2 and led to an extinction coefficient at 868 nm of 71.3, 142.6, or 214 rnM-l cm-l, depending on whether the photosynthetic center was assumed to contain 2, 4, or 6 bacteriochlorophyll molecules. The extinction coefficient of PST,, was determined by mixing photo-oxidized photosynthetic center with ferrocytochrome c in a stopped flow spectrophotometer. Complete reaction was observed when the reactants were mixed in equimolar ratio calculated on the basis of an extinction coefficient of 143 rnr@ cm-l at 868 nm. This photosynthetic center is proposed to contain 4 moles of bacteriochlorophyll, 2 moles of bacteriopheophytin, and 1 mole of spirilloxanthin per equivalent of Pm.

A reaction center – light harvesting 1 complex from a Rhodospirillum rubrum mutant with altered esterifying pigments: characterization by optical spectroscopy and cryo-electron microscopy

Introduction of the bchP gene from Rhodobacter sphaeroides encoding geranylgeranyl reductase into Rhodospirillum rubrum alters the esterification of the bacteriochlorophylls so that phytol is used instead of geranylgeraniol. The resulting transconjugant strain of Rs. rubrum grows photosynthetically, showing that phytolated Bchla can substitute for the native pigment in both the reaction center ...

The reaction center H subunit is not required for high levels of light-harvesting complex 1 in Rhodospirillum rubrum mutants.

The gene (puhA) encoding the H subunit of the reaction center (RC) was deleted by site-directed interposon mutagenesis by using a kanamycin resistance cassette lacking transcriptional terminators to eliminate polar effects in both the wild-type strain Rhodospirillum rubrum S1 and the carotenoid-less strain R. rubrum G9. The puhA interposon mutants were incapable of photoheterotrophic growth but...

Thin-layer chromatography of pigments from reaction center particles of Rhodopseudomonas spheroides.

During the past few years a number of papers have been published, dealing with the isolation of photochemically active reaction center particles from photosynthetic bacteria (e.g. refs 1-6). In these particles the light-harvesting bacteriochlorophyll and most of the other accessory pigments such as carotenoids, have been removed. The particles still contain specialized bactefiochlorophylls, P8o...

Characterization of a Rhodospirillum rubrum plasmid: loss of photosynthetic growth in plasmidless strains.

A single plasmid of 55 kilobases was found in crude cell lysates of each of nine strains of Rhodospirillum rubrum. Restriction endonuclease analysis showed identical fragment patterns with a given nuclease for all plasmids except one, for which an additional EcoRI site was observed. Elimination of the plasmid required that the cells be passaged several times in 25 mM calcium-containing medium, ...
